Proposed working model. Gestational environment enrichment (GEE) in mothers (F0) upregulates IGF1/IGF1R in offspring (F1, embryo and the adults), which in turn phosphorylates/activates ERK and CaMKIV. The phosphorylated pY172-CaMKIV is translocated to the nuclear fraction where it phosphorylates HAT (CBP), thus inducing hyperacetylation at bdnf promoters and upregulating BDNF expression. BDNF preserves synaptic plasticity and improves AD pathologies, which eventually leads to an improved spatial memory capacity. Blockade of HAT, CaMKIV, ERK, and IGF1R abolishes the beneficial effects of GEE
