Astrocytic originated mitochondria transfer to injured DA neurons in a rotenone-induced PD in vitro model. a Mitochondria in astrocytes were labeled with Mito-tracker (green) before co-culture with DA neurons. 24 h after co-culture, immunostaining showed that astrocytic mitochondria were present in DA neurons. Phalloidin dye (blue) and TH (red) were employed to outline the astrocytes and DA neurons respectively. b The presence of astrocytic mitochondria in CellTrace labeled DA neurons was analyzed by flow cytometry. Astrocytic mitochondria were stained with Mitotracker green separately and neurons were labeled with CellTrace (violet) before co-culture. The efficient transfer of Mitochondria Tracker Green labeled mitochondria was confirmed in co-cultures by the presence of dual-positive cells is composed of Mitotracker and CellTrace analyzed by flow cytometry.
