Inhibitory effect of bvPLA2 on genetically induced neuroinflammation in Tg2576 mice. Immunostaining of GFAP, IBA-1, iNOS, and COX-2 in the hippocampus were performed in 20 μm-thick sections of mice brain with specific primary antibodies and the biotinylated secondary antibodies (a). GFAP, IBA-1, iNOS, and COX-2 stainings were quantified by counting the number of positive cells in the field (b). The expression of iNOS, COX-2, GFAP and IBA-1 were detected by Western blotting using specific antibodies in the mice brain (c). Each blot is representative for three experiments. For the cropped images, samples were run in the same gels under the same experimental conditions and processed in parallel. Band density was quantified from three mice (c). The mRNA expression level of pro-inflammatory cytokines (TNF-α, IL-1β, and IL-6) and anti-inflammatory cytokines (IL-4, IL-10, and TGF-β) in the mice brain hippocampus site were assessed by qRT-PCR (d).
